This medication has been associated with considerable cost savings in the hospital setting. For example, a study conducted by Namachivayam et al. found that patients receiving Sildenafil had less time in mechanical ventilation than those treated with a placebo. Moreover, the total length of stay in the intensive care unit was reduced from 189 hours to 47.8%.
A small but significant reduction in pulmonary arterial blood pressure has been demonstrated with Sildenafil in a phase I study of patients with severe chronic thromboembolic PH (PHP). Patients were treated with Sildenafil orally three times a day for two weeks. The study also included a washout period of at least six days between treatment periods. The patients were evaluated for their baseline mPAP, PVRI, and CI. The pulmonary artery blood pressure reductions were statistically significant for all three parameters. The treatment was associated with no adverse events.
Patients should consult their doctor before taking Sildenafil. They should shake the pill thoroughly before taking each dose, shake it well after taking the medication, and make sure they take the prescribed dosage. In addition, they should avoid mixing Sildenafil with other medicines. The pill should also be taken regularly and at the same time every day.
A recent clinical trial evaluated the effects of Fildena in children with pulmonary hypertension. In this trial, Fildena reduced mPAP from 60 mmHg to 50 mmHg. The pulmonary vascular resistance index (PVR) also decreased. This study also found that Sildenafil reduced the PVRI more effectively than NO alone. Furthermore, the patients’ shunt fraction increased from 16.5% to 25. None of the children reported any symptoms of hypoxia during the trial.
